Acceptable microorganisms concentration in a semen sample for in vitro embryo production BJM

The aim of this study was to report that the acceptable concentration of microorganisms in a semen sample for insemination may not be safe for an in vitro fertilization procedure. It seems that the semen sample should be completely germ-free, because of the excellent microorganism proliferation condition promoted by the in vitro environment.

Aspects of energetic substrate metabolism of in vitro and in vivo bovine embryos BJMBR

Although the metabolism of early bovine embryos has not been fully elucidated, several publications have addressed this important issue to improve culture conditions for cattle reproductive biotechnologies, with the ultimate goal of producing in vitro embryos similar in quality to those developing in vivo. Here, we review general aspects of bovine embryo metabolism in vitro and in vivo, and discuss the use of metabolic analysis of embryos produced in vitro to assess viability and predict a viable pregnancy after transference to the female tract.

This study evaluated the influence of follicular fluid (FF) added to the maturation medium on the quality of bovine embryos produced in vitro. The effects of insulin-like growth factor-I (IGF-I) on in vitro maturation (IVM) (experiment I) and on in vitro embryo development (experiment II) of bovine oocytes fertilized in vitro, were evaluated in terms of cleavage (CR), blastocyst (BR) and hatching (HR) rates. For IVM, immature cumulus-oocyte complexes were cultured in TCM-199 medium supplemented with Hepes, sodium bicarbonate, sodium pyruvate, additives, fetal calf serum (B-199 medium) and gonadotropins (14 U/ml PMSG and 7 U/ml hCG). The objective of the present study was to examine the association between follicular fluid (FF) steroid concentration and oocyte maturity and fertilization rates. Seventeen infertile patients were submitted to ovulation induction with urinary human follicle-stimulating hormone, human menopausal gonadotropin and human chorionic gonadotropin (hCG). A total of 107 follicles were aspirated after hCG administration, the oocytes were analyzed for maturity and 81 of them were incubated and inseminated in vitro. Melatonin (MEL) acts as a powerful scavenger of free radicals and direct gonadal responses to melatonin have been reported in the literature. Few studies, however, have evaluated the effect of MEL during in vitro maturation (IVM) on bovine embryos. This study tested the addition of MEL to maturation medium (MM) with no gonadotropins on nuclear maturation and embryo development rates and the incidence of DNA damage in resulting embryos. Cumulus-oocyte complexes were aspirated from abattoir ovaries and cultured in MM (TCM-199 medium supplemented with 10% fetal calf serum - FCS) at 39ºC and 5% CO2 in air. The effects of selenium (Se) in Jersey cows' diet on the aspiration of oocytes and production of embryos in vitro were studied. Groups with five Jersey cows received 3.2mg or 9.6mg Se daily, provided in the feed concentrate. Six follicular aspirations were carried out every 15 days, using only the last 5. The oocytes were classified, and standard procedures were carried out for maturation, fertilization and cultivation. The total number of oocytes (35.11±2.65 vs 23.10±2.16) and degree 1 oocytes (11.61±2.65 vs 4.75±0.97) were higher in the group that received 9.6mg Se and the quantity of naked oocytes (3.23±0.87 vs 6.22±1.18) was lower in this group. A simulation model implemented in the programming software Delphi XE® was applied to evaluate sex selection in bovine. The hypothesis under investigation was that a dynamic model with stochastic and deterministic elements could detect the sexed semen technique to minimize pregnancy cost and to determine the adequate number of recipients required for in vivo (ET) and in vitro embryo production (IVP) in the proposed scenarios. In addition to initial sperm quality, numerous steps can affect the final result of fertilization, from the sperm collection process until gamete mixing (or co-incubation) when spermatozoon enters or fuses with the oocyte. In this review, we summarize the whole process of sperm handling, from collection until fertilization for fish, penaeid shrimp, bivalve mollusks and marine mammals. The study was conducted to determine effect of EB or CS supplements in LAMP reaction on greater accuracy of sexing cattle embryos. Bovine ovaries were collected from the slaughterhouse, oocytes were aspirated from follicles then were evaluated morphology. Oocytes were cultured in TCM-199 at 38.5 ºC 24 h. For in vitro fertilization (IVF), oocytes were co-incubated with spermatozoa at 38.5 ºC 24 h. Fertilization rate was evaluated and then the embryos were in vitro cultured until morula stage. Then, embryo samples were frozen and stored at -196 ºC. Frozen embryos (n=35) were randomly extracted for total cellular DNA and amplified by PCR method. DNA samples were stored at -20 ºC.
